Integrated multi-omic approach reveals the effect of a Graminaceae-derived biostimulant and its lighter fraction on salt-stressed lettuce plants

Plant biostimulants are widely applied in agriculture for their ability to improve plant fitness. In the present work, the impact of Graminaceae-derived protein hydrolysate (P) and its lighter molecular fraction F3 (< 1 kDa) on lettuce plants, subjected to either no salt or high salt conditions, was investigated through the combination of metabolomics and transcriptomics. The results showed that both treatments significantly modulated the transcriptome and metabolome of plants under salinity stress, highlighting an induction of the hormonal response. Nevertheless, P and F3 also displayed several peculiarities. F3 specifically modulated the response to ethylene and MAPK signaling pathway, whereas P treatment induced a down-accumulation of secondary metabolites, albeit genes controlling the biosynthesis of osmoprotectants and antioxidants were up-regulated. Moreover, according with the auxin response modulation, P promoted cell wall biogenesis and plasticity in salt-stressed plants. Notably, our data also outlined an epigenetic control of gene expression induced by P treatment. Contrarily, experimental data are just partially in agreement when not stressed plants, treated with P or F3, were considered. Indeed, the reduced accumulation of secondary metabolites and the analyses of hormone pathways modulation would suggest a preferential allocation of resources towards growth, that is not coherent with the down-regulation of the photosynthetic machinery, the CO2 assimilation rate and leaves biomass. In conclusion, our data demonstrate that, although they might activate different mechanisms, both the P and F3 can result in similar benefits, as far as the accumulation of protective osmolytes and the enhanced tolerance to oxidative stress are concerned. Notably, the F3 fraction exhibits slightly greater growth promotion effects under high salt conditions. Most importantly, this research further corroborates that biostimulants’ mode of action is dependent on plants’ physiological status and their composition, underscoring the importance of investigating the bioactivity of the different molecular components to design tailored applications for the agricultural practice.

Integrated metabolomic and metagenomic strategies shed light on interactions among planting environments, rhizosphere microbiota, and metabolites of tobacco in Yunnan, China

Changes in climatic factors and rhizosphere microbiota led plants to adjust their metabolic strategies for survival under adverse environmental conditions. Changes in plant metabolites can mediate crop growth and development and interact with rhizosphere microbiota of the plant rhizosphere. To understand the interactions among environmental factors, rhizosphere microbiota, and metabolites of tobacco, a study was conducted by using integrated metagenomic and metabolomic strategies at four typical representative tobacco planting sites in Yunnan, China. The results showed that the agronomical and biochemical traits were significantly affected by temperature, precipitation (PREP), soil pH, and altitude. Correlation analyses revealed a significant positive correlation of temperature with length, width, and area of the leaf, while PREP correlated with plant height and effective leaf numbers. Furthermore, total sugar and reducing sugar contents of baked leaves were significantly higher, while the total nitrogen and total alkaloid levels were lower in tobacco leaves at site with low PREP. A total of 770 metabolites were detected with the highest number of different abundant metabolites (DMs) at Chuxiong (CX) with low PREP as compared to the other three sites, in which secondary metabolites were more abundant in both leaves and roots of tobacco. A total of 8,479 species, belonging to 2,094 genera with 420 individual bins (including 13 higher-quality bins) harboring 851,209 CDSs were detected. The phyla levels of microorganisms such as Euryarchaeota, Myxococcota, and Deinococcota were significantly enriched at the CX site, while Pseudomonadota was enriched at the high-temperature site with good PREP. The correlation analyses showed that the metabolic compounds in low-PREP site samples were positively correlated with Diaminobutyricimonas, Nissabacter, Alloactinosynnema, and Catellatospora and negatively correlated with Amniculibacterium, Nordella, Noviherbaspirillum, and Limnobacter, suggesting that the recruitment of Diaminobutyricimonas, Nissabacter, Alloactinosynnema, and Catellatospora in the rhizosphere induces the production and accumulation of secondary metabolites (SMs) (e.g., nitrogen compounds, terpenoids, and phenolics) for increasing drought tolerance with an unknown mechanism. The results of this study may promote the production and application of microbial fertilizers and agents such as Diaminobutyricimonas and Alloactinosynnema to assemble synthetic microbiota community or using their gene resources for better cultivation of tobacco as well as other crops in drought environments.

PLS-based integration of metabolomics and transcriptomics to reveal the effect of different growth years on Eurycoma longifolia roots and hairy root cultures

Growth years are important factors that affect the accumulation of secondary metabolites in Eurycoma longifolia roots. This study aimed to investigate the effect of different growth years on the metabolites and gene expression of E. longifolia roots and hairy root cultures. To achieve this, metabolite profiles and candidate transcripts of terpenoid backbone biosynthesis and sesquiterpenoid and triterpenoid pathways of 1- and 11-year old E. longifolia roots and hairy root cultures were obtained and integrated using partial least squares (PLS). Integration of eighty-two metabolites and forty-five selected candidate genes using PLS successfully differentiated 1-, 11-year-old E. longifolia roots and hairy root cultures, indicating significant metabolite-gene expression differences in response to different growth years and root types. Amino acids, 2-C-methyl-D-erythritol-4-phosphate cytidylyl-transferase (MCT) and acetyl-CoA C-acetyltransferase (AACT) were responsible for the discrimination of 1-year-old E. longifolia root. Quassinoids of eurycomanone and eurycomalactone and genes linked to the upstream synthesis in mevalonate (MVA) pathway contributed to the discrimination of the11-year-old E. longifolia root. 11-hydroxycanthin-6-one and genes that participate in the methylerythritol phosphate (MEP) pathway upstream of terpenoid biosynthesis were responsible for the discrimination of E. longifolia hairy root cultures.

Influence of Cold Stress on Physiological and Phytochemical Characteristics and Secondary Metabolite Accumulation in Microclones of Juglans regia L.

The current study investigated the impact of cold stress on the morphological, physiological, and phytochemical properties of Juglans regia L. (J. regia) using in vitro microclone cultures. The study revealed significant stress-induced changes in the production of secondary antioxidant metabolites. According to gas chromatography-mass spectrometry (GC-MS) analyses, the stress conditions profoundly altered the metabolism of J. regia microclones. Although the overall spectrum of metabolites was reduced, the production of key secondary antioxidant metabolites significantly increased. Notably, there was a sevenfold (7×) increase in juglone concentration. These findings are crucial for advancing walnut metabolomics and enhancing our understanding of plant responses to abiotic stress factors. Additionally, study results aid in identifying the role of individual metabolites in these processes, which is essential for developing strategies to improve plant resilience and tolerance to adverse conditions.

Effects of light-emitting diodes on the morphology and accumulation of glucosinolates, carotenoids and phenolic acids in red kale sprouts

Kale (Brassica oleracea var. acephala) has gained popularity as a nutritious and phytochemical-rich vegetable. This study has investigated the effects of three LED treatments (white, blue, and red) on the accumulation of secondary metabolites in kale sprouts. Ten DAS, the kale sprouts were harvested, and growth measurements were measured. Furthermore, selected sprouts were stored at -80 °C for further biochemical analysis, namely, phenolic acids, glucosinolates, and carotenoids. Sprouts irradiated with red LED light showed the best SL, RT, and FW values. For total GSLs, we found that kale sprouts irradiated with white LED lights showed the best results (41.59±0.41 μmol/g DW). Among the aliphatic GSLs, we found that progoitrin presented the best results under blue LED light (17.93±0.49 µmol/g DW), and among the indolic GSLs, glucobrassicin showed the best results under white and red LED light. The highest concentration of total carotenoids was found in kale sprouts under white LED light exposure (3341.27±206.96 μg/g DW). Individually, β-carotene was observed in high concentration under white LED light (1935.13±87.21 μg/g DW). Among the PAs, chlorogenic acid was found in the highest concentration in the treatments under white LED light (45.78±0.73 µg/g DW). In general, kale sprouts irradiated with white LED light showed high contents of GSLs, carotenoids, and PAs. Regarding morphological characteristics, kale sprouts under red LED light showed the most promise. This research offers a valuable approach to enhancing the phytochemicals found in kale sprouting.

Changes in secondary metabolites contents and stress responses in Salvia miltiorrhiza via ScWRKY35 overexpression: Insights from a wild relative Salvia castanea

Salvia castanea Diels, a close wild relative to the medicinal plant, Salvia miltiorrhiza Bunge, primarily grows in high-altitude regions. While the two species share similar active compounds, their content varies significantly. WRKY transcription factors are key proteins, which regulate plant growth, stress response, and secondary metabolism. We identified 46 ScWRKY genes in S. castanea and found that ScWRKY35 was a highly expressed gene associated with secondary metabolites accumulation. This study aimed to explore the role of ScWRKY35 gene in regulating the accumulation of secondary metabolites and its response to UV and cadmium (Cd) exposure in S. miltiorrhiza. It was found that transgenic S. miltiorrhiza hairy roots overexpressing ScWRKY35 displayed upregulated expression of genes related to phenolic acid synthesis, resulting in increased salvianolic acid B (SAB) and rosmarinic acid (RA) contents. Conversely, tanshinone pathway gene expression decreased, leading to lower tanshinone levels. Further, overexpression of ScWRKY35 upregulated Cd transport protein HMA3 in root tissues inducing Cd sequestration. In contrast, the Cd uptake gene NRAMP1 was downregulated, reducing Cd absorption. In response to UV radiation, ScWRKY35 overexpression led to an increase in the accumulation of phenolic acid and tanshinone contents, including upregulation of genes associated with salicylic acid (SA) and jasmonic acid (JA) synthesis. Altogether, these findings highlight the role of ScWRKY35 in enhancing secondary metabolites accumulation, as well as in Cd and UV stress modulation in S. miltiorrhiza, which offers a novel insight into its phytochemistry and provides a new option for the genetic improvement of the plants.

Microbial communities of Schisandra sphenanthera Rehd. et Wils. and the correlations between microbial community and the active secondary metabolites.

Schisandra sphenanthera Rehd. et Wils. is a plant used in traditional Chinese medicine (TCM). However, great differences exist in the content of active secondary metabolites in various parts of S. sphenanthera. Do microorganisms critically influence the accumulation of active components in different parts of S. sphenanthera? In this study, 16S/ITS amplicon sequencing analysis was applied to unravel microbial communities in rhizospheric soil and different parts of wild S. sphenanthera. At the same time, the active secondary metabolites in different parts were detected, and the correlation between the secondary metabolites and microorganisms was analyzed. The major components identified in the essential oils were sesquiterpene and oxygenated sesquiterpenes. The contents of essential oil components in fruit were much higher than that in stem and leaf, and the dominant essential oil components were different in these parts. The dominant components of the three parts were γ-muurolene, δ-cadinol, and trans farnesol (stem); α-cadinol and neoisolongifolene-8-ol (leaf); isosapathulenol, α-santalol, cedrenol, and longiverbenone (fruit). The microbial amplicon sequences were taxonomically grouped into eight (bacteria) and seven (fungi) different phyla. Community diversity and composition analyses showed that different parts of S. sphenanthera had similar and unique microbial communities, and functional prediction analysis showed that the main functions of microorganisms were related to metabolism. Moreover, the accumulation of secondary metabolites in S. sphenanthera was closely related to the microbial community composition, especially bacteria. In endophytic bacteria, Staphylococcus and Hypomicrobium had negative effects on five secondary metabolites, among which γ-muurolene and trans farnesol were the dominant components in the stem. That is, the dominant components in stems were greatly affected by microorganisms. Our results provided a new opportunity to further understand the effects of microorganisms on the active secondary metabolites and provided a basis for further research on the sustainable utilization of S. sphenanthera.

Effects of combined treatment of light quality and melatonin on health-promoting compounds in Chinese kale sprouts

Chinese kale sprouts have garnered attention owing to its rich health-promoting compounds. This study aimed to investigate the effects of different light qualities (white, blue, and red light) and melatonin (100 μmol L−1), both individually and in combination, on the growth and accumulation of health-promoting compounds in Chinese kale sprouts. Red light positively influenced sprout growth (compared with white light, plant height and fresh weight increased by 16.67 % and 4.87 %, respectively), and the red light + melatonin treatment further enhanced growth (compared with single red light, the plant height and fresh weight increased by 14.97 % and 12.29 %, respectively), while increasing glucosinolate accumulation through the up-regulation of glucosinolate biosynthetic genes (UGT74B1, CYP83B1, SOT18 and IGMT1). Conversely, blue light inhibited sprout growth, elevated the expression levels of PAL, CHS, CHI, GGP, and PAX, improved antioxidant levels, and accelerated the accumulation of aliphatic glucosinolates, reducing indolic glucosinolates. The blue light + melatonin treatment promoted the accumulation of sprout biomass and anthocyanins (11.91 % and 13.33 % higher than that of single blue light). The study unveiled various pathways influencing plant growth, development, and secondary metabolite accumulation, including plant circadian rhythms, ascorbate and aldarate metabolism, phenylpropanoid biosynthesis, and glucosinolate biosynthesis.

Zinc-Enhanced Regulation of the Ginkgo biloba L. Response and Secondary Metabolites

Ginkgo biloba L. leaves are rich in secondary metabolites with important medicinal values; to increase their contents, foliar spraying of micronutrients is a potential strategy. Zinc, a multifunctional element, has a significant impact on the content of secondary metabolites in other plants, but relevant research into ginkgo is still lacking. In our study, different spraying time and concentration strategies were used to investigate the effects of zinc sulfate (ZnSO4) on physiological indicators and secondary metabolites of 2-year-old ginkgo. The results demonstrated that ZnSO4 could increase the contents of hydrogen peroxide, abscisic acid, and free amino acids in ginkgo leaves. It also enhances the antioxidant enzyme activity of ginkgo leaves, decreases the content of plant auxin, and ultimately facilitates the accumulation of ginkgo terpene lactones (TTL). Spraying ZnSO4 in June resulted in a more significant increase in the contents of TTL and flavonoids compared to spraying in August. After spraying 12 mmol/L ZnSO4 in June, the contents of TTL and flavonoids in ginkgo leaves were significantly elevated by 35.95% and 24.30%, respectively, compared to those in the CK (p &lt; 0.05). The contents of ginkgolide A, B, and C were notably increased by 45.93%, 46.56%, and 74.29%, respectively, compared to those in the CK (p &lt; 0.05). Therefore, our study suggests that the optimal timing for spraying ZnSO4 on ginkgo is in June, with a recommended concentration of 12 mmol/L. Our study provides a theoretical basis for the accumulation of secondary metabolites in ginkgo and guides the production of its leaf-utilization plantations.

Constructed the metabolic compound accumulation profiles of daylily at different main producing areas using multi-omics techniques

Daylily flower buds (DFB) are widely cultivated in China. The discernible disparities exist in the nutritional composition of daylilies across different primary production areas. However, limited studies about the chemical composition and biosynthetic mechanisms of the principal constituents of DFB. This study systematically investigated daylilies from seven distinct growing regions by integrated metabolomics and transcriptomic sequencing to elucidate the environmental impact on DFB quality and metabolite biosynthesis mechanisms. In DFB, flavonoids constituting the majority, followed by phenolic acids and lipids. The total flavonoid and polyphenol content exhibited the highest levels in northern China (Ningxia, Gansu), and the lowest levels in southern regions (Hunan, Fujian). Correspondingly, antioxidant enzyme activity and free radical scavenging capacity mirrored these regional disparities. Thus, northern-producing areas demonstrated a propitious environment for the accumulation of secondary metabolites, particularly flavonoids and phenolic acids. In contrast, southern production areas exhibited a less conducive environment for the accumulation of flavonoid glycosides in daylilies. Furthermore, distinctive variations in the key metabolites were observed in FJQZ, a region characterized by diverse flower colors. Key enzymes and transcription factors were identified and validated through RT-qPCR. These results contribute valuable insights into the daylily quality formation mechanisms, offering a foundation for high-quality breeding strategies.

Arbuscular Mycorrhizal Fungi-Mediated Modulation of Physiological, Biochemical, and Secondary Metabolite Responses in Hemp (Cannabis sativa L.) under Salt and Drought Stress.

The increasing impact of global climate change has resulted in adversity stresses, like salt and drought, gradually becoming the main factors that limit crop growth. Hemp, which contains numerous medicinal active components and multiple bioactive functions, is widely used in the agricultural, industrial, and medical fields, hence promoting the rapid development of related industries. Arbuscular mycorrhizal fungi (AMF) can establish a symbiotic relationship with 80% of vascular plants. This symbiosis promotes host plant growth, regulates plant physiology and biochemistry, facilitates secondary metabolite synthesis, and enhances resistance to abiotic stresses. However, the effects of salt stress, drought stress, and AMF interaction in hemp are not well understood. In this study, to investigate this, we performed a study where we cultured hemp that was either inoculated or uninoculated with Funneliformis mosseae and determined changes in effective colonization rate, growth, soluble substances, photosynthesis, fluorescence, ions, and secondary metabolites by cultivating hemp under different concentrations of NaCl (0 mM, 100 mM, and 200 mM) and different soil moisture content (45%, 25%, and 15%). The results showed that salt, drought stress, or salt-drought interaction stress all inhibited colonization rate after stress, plant growth, mainly due to ion toxicity and oxidative damage. Inoculation with F. mosseae effectively alleviated plant growth inhibition under 100 mM NaCl salt stress, drought stress, and salt-drought interaction stress conditions. It also improved osmoregulation, photosynthetic properties, fluorescence properties, and ion homeostasis, and promoted the accumulation of secondary metabolites. However, under 200 mM NaCl salt stress conditions, inoculation with F. mosseae negatively affected plant physiology, biochemistry, and secondary metabolite synthesis, although it did alleviate growth inhibition. The results demonstrate that there are different effects of salt-drought interaction stress versus single stress (salt or drought stress) on plant growth physiology. In addition, we provide new insights about the positive effects of AMF on host plants under such stress conditions and the effects of AMF on plants under high salt stress.

Current Advances in the Functional Diversity and Mechanisms Underlying Endophyte-Plant Interactions.

Plant phenotype is a complex entity largely controlled by the genotype and various environmental factors. Importantly, co-evolution has allowed plants to coexist with the biotic factors in their surroundings. Recently, plant endophytes as an external plant phenotype, forming part of the complex plethora of the plant microbial assemblage, have gained immense attention from plant scientists. Functionally, endophytes impact the plant in many ways, including increasing nutrient availability, enhancing the ability of plants to cope with both abiotic and biotic stress, and enhancing the accumulation of important plant secondary metabolites. The current state of research has been devoted to evaluating the phenotypic impacts of endophytes on host plants, including their direct influence on plant metabolite accumulation and stress response. However, there is a knowledge gap in how genetic factors influence the interaction of endophytes with host plants, pathogens, and other plant microbial communities, eventually controlling the extended microbial plant phenotype. This review will summarize how host genetic factors can impact the abundance and functional diversity of the endophytic microbial community, how endophytes influence host gene expression, and the host-endophyte-pathogen disease triangle. This information will provide novel insights into how breeders could specifically target the plant-endophyte extended phenotype for crop improvement.

Influence of Abiotic and Biotic Elicitors on Organogenesis, Biomass Accumulation, and Production of Key Secondary Metabolites in Asteraceae Plants.

The medicinal plants of the Asteraceae family are a valuable source of bioactive secondary metabolites, including polyphenols, phenolic acids, flavonoids, acetylenes, sesquiterpene lactones, triterpenes, etc. Under stressful conditions, the plants develop these secondary substances to carry out physiological tasks in plant cells. Secondary Asteraceae metabolites that are of the greatest interest to consumers are artemisinin (an anti-malarial drug from Artemisia annua L.-sweet wormwood), steviol glycosides (an intense sweetener from Stevia rebaudiana Bert.-stevia), caffeic acid derivatives (with a broad spectrum of biological activities synthesized from Echinacea purpurea (L.) Moench-echinacea and Cichorium intybus L.-chicory), helenalin and dihydrohelenalin (anti-inflammatory drug from Arnica montana L.-mountain arnica), parthenolide ("medieval aspirin" from Tanacetum parthenium (L.) Sch.Bip.-feverfew), and silymarin (liver-protective medicine from Silybum marianum (L.) Gaertn.-milk thistle). The necessity to enhance secondary metabolite synthesis has arisen due to the widespread use of these metabolites in numerous industrial sectors. Elicitation is an effective strategy to enhance the production of secondary metabolites in in vitro cultures. Suitable technological platforms for the production of phytochemicals are cell suspension, shoots, and hairy root cultures. Numerous reports describe an enhanced accumulation of desired metabolites after the application of various abiotic and biotic elicitors. Elicitors induce transcriptional changes in biosynthetic genes, leading to the metabolic reprogramming of secondary metabolism and clarifying the mechanism of the synthesis of bioactive compounds. This review summarizes biotechnological investigations concerning the biosynthesis of medicinally essential metabolites in plants of the Asteraceae family after various elicitor treatments.

Overexpression of AtMYB2 Promotes Tolerance to Salt Stress and Accumulations of Tanshinones and Phenolic Acid in Salvia miltiorrhiza.

Salvia miltiorrhiza is a prized traditional Chinese medicinal plant species. Its red storage roots are primarily used for the treatment of cardiovascular and cerebrovascular diseases. In this study, a transcription factor gene AtMYB2 was cloned and introduced into Salvia miltiorrhiza for ectopic expression. Overexpression of AtMYB2 enhanced salt stress resistance in S. miltiorrhiza, leading to a more resilient phenotype in transgenic plants exposed to high-salinity conditions. Physiological experiments have revealed that overexpression of AtMYB2 can decrease the accumulation of reactive oxygen species (ROS) during salt stress, boost the activity of antioxidant enzymes, and mitigate oxidative damage to cell membranes. In addition, overexpression of AtMYB2 promotes the synthesis of tanshinones and phenolic acids by upregulating the expression of biosynthetic pathway genes, resulting in increased levels of these secondary metabolites. In summary, our findings demonstrate that AtMYB2 not only enhances plant tolerance to salt stress, but also increases the accumulation of secondary metabolites in S. miltiorrhiza. Our study lays a solid foundation for uncovering the molecular mechanisms governed by AtMYB2 and holds significant implications for the molecular breeding of high-quality S. miltiorrhiza varieties.

DNA methylation regulates the secondary metabolism of saponins to improve the adaptability of Eleutherococcus senticosus during drought stress

Plant growth and development can be significantly impacted by drought stress. Plants will adjust the synthesis and accumulation of secondary metabolites to improve survival in times of water constraint. Simultaneously, drought stress can lead to modifications in the DNA methylation status of plants, and these modifications can directly impact gene expression and product synthesis by changing the DNA methylation status of functional genes involved in secondary metabolite synthesis. However, further research is needed to fully understand the extent to which DNA methylation modifies the content of secondary metabolites to mediate plants’ responses to drought stress, as well as the underlying mechanisms involved. Our study found that in Eleutherococcus senticosus (E. senticosus), moderate water deprivation significantly decreased DNA methylation levels throughout the genome and at the promoters of EsFPS, EsSS, and EsSE. Transcription factors like EsMYB-r1, previously inhibited by DNA methylation, can re-bind to the EsFPS promotor region following DNA demethylation. This process promotes gene expression and, ultimately, saponin synthesis and accumulation. The increased saponin levels in E. senticosus acted as antioxidants, enhancing the plant’s adaptability to drought stress.

Analysis of light recipe, seeding density, and fertilization effects on secondary metabolite accumulation and growth-defense responses in Brassicaceae microgreens

Although there have been large improvements to crop yield over time, this has not been accompanied by an increase in human nutritional wellbeing. In fact, there are worsening health crises associated with over- and under-consumption of particular food groups, resulting in negative human health outcomes. One solution to this is to utilize controlled environment agriculture to produce microgreens that have a high density of valuable Secondary Metabolites (SMs) such as antioxidants, phenolics, or pigment molecules that are associated with positive human health outcomes. However, optimal growth recipes to produce microgreens and their valuable nutritional compounds are not well known due to much species-specific variation, as well as biological tradeoffs between biomass and SM production. This is known as the growth defense hypothesis which describes how plants have a finite carbon budget from which to allocate to growth, or defensive, processes. To further research in this regard, this project used climate chambers to grow three species of Brassicaceae microgreens (kohlrabi, mustard, and radish) in highly controlled environmental conditions, where we used five Light Recipes, two fertilization levels, and two seeding density treatments. Our results showed that there were significant differences in SM production of these microgreens due to changes in incident light, as well as significant interactions between Fertilizer and Light Recipe for all SMs except Anthocyanins. For example, for all three species, the High Far-Red Light Recipe had the significantly highest Phenolic concentration, but with lower values of the other four SMs. The low-intensity 24-volt high efficiency LEDs had the significantly highest Trolox Equivalent Antioxidant Capacity (TEAC) concentrations, while the High Red recipe had the highest Ferric Reducing Antioxidant Power (FRAP) and Flavonoid concentrations. For Anthocyanins, there were less clear patterns, with the No Green or High Red recipes having generally higher concentrations, but not always significantly. We did find some evidence supporting the growth-defense hypothesis, where our higher biomass values were negatively correlated with SM concentrations. We also found significant differences between the concentrations of SMs in leaves and stems for kohlrabi and mustard microgreens. Finally, we found some significant relationships between increasing Fertilizer dosage and SMs, for instance that Flavonoids and FRAP concentrations increased with Fertilizer application, while Anthocyanin decreased, and Phenolic and TEAC had little effect. In conclusion, we found that there were significant relationships for Light Recipe and Fertilizer, and oftentimes their interaction, for the accumulation of SMs in Brassicaceae microgreens, which can inform microgreen production environments.

NaCl elicitation enhances metabolite accumulation and stress resilience in Inula crithmoides L. shoot cultures: implications for its nutritional and medicinal value

This study explored the impact of sodium chloride (NaCl) elicitation on the accumulation of primary and secondary metabolites and the oxidative stress responses of Inula crithmoides L. (golden samphire) in vitro shoot cultures. Elicitation involved applying different concentrations of NaCl (0, 50, 100, and 200 mM) for 4 weeks. This was followed by assessing its impact on plant growth, physiological parameters (pigments, hydrogen peroxide content, total soluble sugars and proteins, and proline), and secondary metabolism (phenylalanine ammonia-lyase activity, shikimic acid, phenolics, flavonoids, and hydroxycinnamic acids) in the shoots. The extracts were also analysed using high-performance liquid chromatography (HPLC). The NaCl elicitation did not affect shoot growth but increased physiological functions such as photosynthesis and oxidative stress management under moderate salinity levels. In addition, NaCl treatments increased the synthesis of soluble sugars and proteins, particularly proline, as well as bioactive phenolics such as gentisic acid, chlorogenic acid, 4-hydroxybenzoic acid, luteolin-7-O-glucoside, and naringenin-7-O-glucoside. The NaCl elicitation in golden samphire shoot cultures offers a significant method for enhancing the production of important nutritional and bioactive compounds. This underscores the species’ potential for cultivation in saline environments and provides valuable prospects for its utilization in the health and nutrition sectors.

ФИЗИОЛОГИЧЕСКИЕ ОСОБЕННОСТИ СОРТОВ ГРУШИ НА ТЕРРИТОРИИ КРАСНОДАРСКОГО КРАЯ

The search for new promising pear varieties with high adaptation and production potential is an important aim of modern breeding for the southern regions of the Russian Federation. In present study, the results of the physiological characteristics of four pear varieties during the summerautumn season 2022-2023 are presented. Pear trees were planted in the territory of the Prikubanskaya fruit growing in the Krasnodar Region. Three varieties Leven, Lyuberskaya, Flamenco of Russian breeding and the American variety Devo were used. The pear leaves were selected three times during the summer months and in September. The content of soluble sugars and total phenols, as well as the activity of antioxidant enzymes – superoxide dismutase and peroxidase – were determined in the plant material. It was established that an increase of the level of the primary and secondary metabolites accumulation and an increase of enzymatic activity by ~20-75 % occurred in the hotter and drier 2023. The content of soluble sugars significantly increased to ~12.0 mg/g fresh weight in the leaves of the varieties Devo and Leven. The accumulation of phenolic compounds increased in all pear varieties by more than 30 % with values exceeding 27,0 mg/g fresh weight. The level of superoxide dismutase activity in 2023 averaged 13.4 unit/mg protein, which is slightly higher than the values of 2022. Peroxidase activity increased 2-4 times with maximum values of more than 8,0 unit/mg protein per min. in the varieties Leven and Lyuberskaya. In conclusion, the data obtained indicate that the drier and hotter conditions of 2023 were not critical for the normal growth and development of the studied pears varieties, but only initiated the activation of the defense antioxidant system in plant cells. Identification and Functional Analysis of 1-Deoxy-D-xylulose-5-phosphate Synthase Gene in Tomatoes (Solanum lycopersicum)

1-deoxy-D-xylulose-5-phosphate synthase (DXS) is a rate-limiting enzyme in terpene synthesis that can affect the accumulation of secondary metabolites in plants. In this study, three DXS gene family members were identified in the tomato genome-wide database. Using bioinformatics methods, we analyzed the gene structure, evolutionary affinities, and cis-acting elements of the SlDXS gene family members. Promoters of SlDXS genes contain plant hormone-responsive elements such as the CGTCA-motif, TGACG-motif, ABRE, TCA-element, TGA-element, ERE, CAT-box, and AACA-motif, which suggested that the SlDXS gene family may play an important role in hormone response. The RT-qPCR analysis showed that the tomato DXS2 gene was able to respond upon exposure to methyl jasmonate (MeJA). The construction of a virus-induced gene silencing (VIGS) vector for the SlDXS gene showed that the SlDXS2 gene was also able to respond to MeJA in silenced plants, but the induction level was lower relative to that of wild-type plants. The SlDXS1 gene is associated with the synthesis of photosynthetic pigments. This study provides a reference for the further elucidation of the DXS gene’s biological function in the terpenoid synthesis pathway in tomatoes.

Foliar phenols and flavonoids level in pteridophytes: an insight to culturable fungal endophyte colonisation.

There are many available reports of secondary metabolites as bioactive molecules from culturable endophytes, nevertheless, there are scarce research pertaining to the levels of metabolites in plants with respect to the incidence and colonisation of fungal endophytes in the same foliar tissues. Therefore, the study was focussed to examine whether fungal endophyte colonisation and the accumulation of secondary metabolites, such as flavonoids and phenols, in the plants are related in any way. For this reason, the study aims to analyse phenols and flavonoids from the fronds of eleven pteridophytes along with the culture-dependent isolation of fungal endophytes from the host plants subsequently assigning them to morphological category and their quantitative analysis and further resolving its identities through molecular affiliation. The results revealed that nine morpho-categories of fungal endophytes were allotted based on culture attributes, hyphal patterns and reproductive structural characters. Highest numbers of species were isolated from Adiantum capillus-veneris and least was recorded from Pteris vittata and Dicranopteris linearis. Maximum phenol content was analysed from the fronds of P. vittata and lowest was recorded in A. capillus-veneris. Highest flavonoid content was measured in D. linearis and lowest was detected in Christella dentata. Significant negative correlation was observed between phenol content of ferns and species richness of fungi. Moreover, significant positive correlation was observed with the relative abundance of Chaetomium globosum and flavonoid content of ferns and negative significant relation was found between relative abundance of Pseudopestalotiopsis chinensis and phenol content of pteridophytes. The occurrence and the quantitative aspects of endophytes in ferns and their secondary metabolites are discussed.
